1H-imidazole,5,5'-[1,1' -biphenyl]-4,4' -bis [2-(2S) -2-pyrrolidine hydrochloride (1:4) is an important intermediate in the production of dactavir, a hepatitis C virus (HCV) NS5A inhibitor for genotype 3 infection for the treatment of chronic hepatitis C infection. On July 24, 2015, the FDA approved Squibb's chronic hepatitis C drug Daclatasvir-Dactamivir. Squibb's application for FDA approval of dactamivir was also torturously rejected by the FDA, but in mid-2015, the FDA approved the combination of dactamivir and sofibuvir for patients with hepatitis C genotype 3. Chemicalbook was approved in Japan, the European Union and South Korea before FDA approval. In 2014, the Japanese Ministry of Health approved dactamivir with asuChemicalbooknaprevir (Sunvepra) for HCV 1 infection. The European Union also approved Daclatasvir in combination with other agents for hepatitis C gene types 1,2,3 and 4 in 2014. Daclatasvir is the first NS5A complex inhibitor approved in the European Union (EU) for "shorter treatment duration (12 or 24 weeks)" compared to 48 weeks of treatment with interferon and ribavirin based regimens. Daclatasvir single therapy is not recommended. At present, daclatasvir combined therapy with Sofebvir is the mainstream regimen, which is characterized by good efficacy, higher SVR, fewer side effects, and shorter treatment period than other regimen
